EIN3-binding F-box proteins identified from petunia corollas using RNA-seq

Recent advances in RNA-sequencing technologies have greatly enhanced the ability to generate sequence data and transcriptome profiles. RNA-seq was utilized in Petunia × hybrida 'Mitchell Diploid' to identify gene changes in corollas after pollination. More than 4,700 genes were differentially expressed in the corolla between 12 and 24 h after pollination (hap). Many of these genes were involved in hormone synthesis or signaling. A putative EIN3-binding F-box protein (EBF) was identified among the differentially expressed genes. A reciprocal BLASTx revealed three putative PhEBFs in the petunia RNA-seq dataset. PhEBF1b and PhEBF2 had predicted F-box domains, but the translated PhEBF1a did not have an F-box domain (pfam00646) that met the statistical threshold for the Conserved Domain Database (CDD). PhEBF1a and PhEBF1b expression was at least 4-fold higher in pollinated corollas compared to unpollinated corollas at the same time points. PhEBF2 mRNA was not up-regulated by pollination as much as PhEBF1a and 1b, and mRNA abundance decreased by 48 h after pollination when corollas were visibly wilted.
