- We describe a method for sample size calculation in a pooling design to determine the content of a substance in food. This can be used to calculate the pool size and the number of measurements needed to estimate the mean within a pre-specified confidence interval width. The effect of pooling on the decrease of measurements needed depends on the ratio between the biological variance (variation between different samples) and the technical variance (variation between repeated measures of one sample). The application of the presented method is shown for measuring dioxins and polychlorinated biphenyls (PCB) in eggs. It can, however, be transferred to other contaminants and foods as well.
